I'm having some difficulty understanding why 18 is C in the Examkrackers Reasoning Lecture ii ICE. The western blot seems like the WT group has the 29 kDa and 35 kDa bands but the answers say it only has the 35 kDa. Could someone please explain how you are supposed to read the western blot diagrams?

Hi Pathik!

Remember that the darker the smear, the more protein (in this case Aqp2) is present. You can see that there is a prominently dark streak at the 31 kDA form on the 3rd panel, and not particularly prevalent in the other two. This allows us to rule out A and B as an answer. Choice C tells us that 31 kDA form is present in the third panel (which we know to be true) and that 35-45kDA is expressed in the wild type, which you can see as the darkest smear on the first panel. While it is true that 29 may be present in the WT group, that is not one of the answers, and everything in C is correct so that is the best choice.
